Expression and Significance of Cox-2 and Ang-2 in Colorectal ...

[Abstract]

Objective: To study the expression of cyclooxygenase-2 (cox-2) and angiopoietin-2 (Ang-2) in colorectal cancer tissue, tissue adjacent to the cancer, normal tissue and the relationship between these expressions and the clinic pathological parameters of colorectal cancer. And to investigate the effect on cell growth, expression of VEGF and Angiopointin-2 protein in HT-29 cell line with the treatment of Meloxicam, a selected NSAIDs.Methods: Immunohistochemical SP technique method was used to detect the expression of cox-2 and Ang-2 protein in 45 colon cancer tissue, 45 tissue adjacent to the cancer and 15 normal tissue. Then cultured HT-29 cells were treated by Meloxicam with various concentration and time. Cell Counting Kit-8 (CCK-8) detected the proliferation of HT-29. The levels of VEGF and Ang-2 protein in supernatants were determined by enzyme-linked immunosorbent assay (ELISA). The expression of VEGF and Ang-2 mRNA in cultured HT-29 were determined by real-time quantitative reverse- transcription-polymerase chain reaction.Results: cox-2 and Ang-2 were expressed in tumor tissue and tissue adjacent to tumor, were not expressed in normal tissue. In 45 colon cancer tissue, the positive expression ratio of cox-2 and Ang-2 were 80.0%, 66.7%respectively; in 45 tissue adjacent to the cancer, the positive expression rates of cox-2 and Ang-2 were 35.6%, 11.1%. There was a close correlationship between the expressions of cox-2 and Ang-2 with several clinic pathological factors including lymph node metastasis and Dukes stage. And there was a close correlation between the expression of cox-2 and Ang-2. And there was a cytotoxicity in cell proliferation of HT-29 with Meloxicam treatment in different concentration and time, a concentration-effect and time- effect relationship was showed to some extent. Cell cycle analysis showed that the cells were mainly blocked in G0/G1phase.The VEGF and Ang-2 protein level in supernatants was reduced gradually with the concentration or time increasing; there was a dependence on the concentration and action time. The expression of cox-2?VEGF and Ang-2mRNA was reduced gradually with the concentration increasing, and also there was a dependence on the concentration.Conclusion: cox-2 and Ang-2 play an important role in the happen and development of colon cancer..Meloxicam can reduce the expression of VEGF and Ang-2 at the level of protein and mRNA in colon carcinoma cell line. Application of specific inhibitor of cox-2 in clinic is significative.
